AAV-based intersectional targeting

Our growing understanding of cell diversity generates an increasing need for molecular tools that selectively target cells on the basis of multiple features. Hughes et al. present an AAV-based toolkit that uses hammerhead ribozymes to achieve intersectional transgene expression. In the cover image, overlapping vinyl records form an abstract representation of Venn diagrams that highlight the intersectionality of complex features.

  • Effective science communication is necessary for engaging the public in scientific discourse and ensuring equitable access to knowledge. Training doctoral students in science communication will instill principles of accessibility, accountability, and adaptability in the next generation of scientific leaders, who are poised to expand science’s reach, generate public support for research funding, and counter misinformation. To this aim, we provide a guide for implementing formal science communication training for doctoral students.

  • After spinal cord injury, stromal fibroblasts originate from pericytes and perivascular fibroblasts, with pericytes more prevalent in gray matter and fibroblasts in white matter. Holl et al. show that both cell types respond to injury and inflammation, are activated, and transcriptionally converge on scar formation after injury, paving the way for therapeutic possibilities.

  • We present a developmental atlas that offers insight into sequential epigenetic changes underlying early human brain development modeled in organoids, which reconstructs the differentiation trajectories of all major CNS regions. It shows that epigenetic regulation via the installation of activating histone marks precedes activation of groups of neuronal genes.
