In an ex vivo skin explant model, the number of Langerhans cells (LCs) migrating from individual explants was not significantly affected by prior exposure to HSV-2 or HIV-1. However, pre-incubation of skin explants with HSV-2 before exposure to HIV-1 significantly increased (by a factor of three) the percentage of emigrating LCs that were HIV+ compared with exposure to HIV-1 alone. Emigrating LCs co-infected with HSV-2 and HIV-1 were rarely detected, which indicates that this is unlikely to be a direct effect. Instead, supernatants from HSV-2-infected epithelial cell cultures could increase the percentage of HIV+ monocyte-derived LCs (mLCs) after exposure to HIV in culture in a dose-dependent manner, which points to an indirect mechanism for increasing HIV susceptibility.
