Abstract
THE staining of cell cultures infected with viruses by fluorescein-labelled antibodies has become almost a routine technique in virus diagnostic and research laboratories. Tissue culture cells are usually grown and infected on floating or Leighton coverslips. At the proper time the coverslips are washed and the cells are fixed with acetone or some other suitable fixative. The coverslips are then supported in a moist chamber with the cells uppermost and are overlaid with fluorescein-labelled antibody solution. After staining for various lengths of time the coverslips are washed, mounted and observed under the ultra-violet light microscope.
