A Direct Microbiological Assay for Urine Phenylalanine in Phenylketonuric Screening

Abstract

INCREASED amounts of both phenylalanine and ortho-hydroxyphenylacetic acid are found in the urine of phenylketonuric (PKU) infants at age 5–8 days, frequently several days before phenylpyruvic acid can be detected in the urine1. Determination of these substances in the urine by paper chromatography is not suited to large-scale screening programmes. The direct microbiological assay procedure described here is simple enough to be used as a screening test.
