Reversible Reaction of Double-Stranded Polynucleotides and HgII : Separation of the Strands

Abstract

THE reaction of double-stranded deoxyribonucleic acids (DNA) and mercuric compounds1 proceeds by two principal steps2–4. In the DNA's occurring naturally, these steps can be demonstrated and defined by the presence of isobestic points in the ultraviolet spectra of the equilibrium mixture. The first reaction occurs in the region 0 r 0.5 – 0.6; the second, in the region r 0.5 – 0.6, where, as usual, the quantity r is defined as the number of moles of HgII added per mole of nucleotide.
