Abstract
N-methyl-N-nitro-N-nitrosoguanidine (nitrosoguanidine) is a powerful and widely used mutagen. Using synchronized populations of Escherichia coliwe have shown that large numbers of mutations occur at specific loci when these loci are replicated1. This specificity makes it possible to use nitrosoguanidine to direct mutagenesis, to study the mode of replication of the chromosome and to map the chromosome1–4.
