Abstract
STUDIES on conditionally lethal mutants of polyoma virus or simian virus 40 (SV40) should provide information on viral gene functions in infected cells. Temperature-sensitive mutants of polyoma virus1–4 and SV40 (refs 5–8) and a host range mutant of polyoma virus9,10 have already been isolated. But, the functional relationship between the viral genes of polyoma virus and SV40 has not yet been studied. To examine the relationship between the viral gene functions of polyoma virus and SV40, and changes of the cellular regulation system in cells transformed by SV40, we attempted to isolate new host range mutants of polyoma virus which could replicate in mouse cells transformed by SV40 but not in normal mouse cells. We report here the isolation of two host range mutants of this type. Productive infection of these mutants in A31-714 cells11 (subclone of mouse BALB/3T3), which are restrictive for these mutants, is facilitated by co-infection with SV40.
