Di- and trimethylated congeners of 7-methylguanine in Sindbis virus mRNA

Abstract

IN the past two years it has become clear that most animal messenger RNA—both cellular1–6 and viral7–11—is “capped” by 7-methylguanosine (m7G) residues present in inverted nucleotide linkage at 5′ termini of the general form m7G (5′)pppNm. At least for certain mRNAs the capping m7G has an important role in mRNA-ribosome binding and thus in initiation of protein synthesis12–15. We now report novel forms of capped 5′ termini on the main mRNA, ‘26S’ RNA, coded for by the animal alphavirus, Sindbis. These termini contain, in addition to the usual m7G, its hypermodified congeners m22,7G and m32,2,7G. This last compound has been previously described only in small nuclear (sn) RNAs of unknown function16,17, whereas m22,7G has not been described previously.
