Abstract
RNA polymerase lacking σ-factor can transcribe the replicative form of phage fd DNA quite well, but loses the ability to select the appropriate initiation sites on the template, and both strands are transcribed. When σ-factor is added, the specificity and asymmetry of transcription are restored. The σ-factor restricts initiation to specific sites on the DNA template.
