Culture of Rabbit Embryo in Circulating Medium

Abstract

OTHER reports from this laboratory have described the growth of late preimplantation to postimplantation stage rabbit embryos in vitro, in open culture dishes containing static medium1,2, and in large volumes of media with continuous bubbling of gases3. (Various types of roller tube cultures have also been attempted, but results have not been encouraging.) This article reports the stage of growth achieved when rabbit embryos are cultured in recirculating medium using a modification of the system designed by New4 for postimplantation rodent embryos. The earliest stages that have been successfully grown by this method are the 8-day post coitum hamster5 and 7.5 day post coitum rat embryos6. In these cases, the embryos at explantation were at the egg-cylinder to late pre-somite stages. In the work reported here using the rabbit, embryos were explanted 6 days and 18 h post coitum (at least 6 h before implantation) while still at early or preprimitive streak stages.
