Factor producing Separable Strands during Replication of a Synthetic DNA

Abstract

REPORTS from this laboratory have described the replication of synthetic, defined DNA-like polymers consisting of repeating mono1, di1,2, tri3, and tetra nucleotide4 sequences. The enzyme used was fraction VII DNA polymerase5, prepared in the laboratory of A. Kornberg. Both of the complementary templates were necessary to obtain many-fold syntheses and both strands were replicated in equal amounts. In the cases of dAn : dTn and d(TG)n : (CA)n, the products could be separated into their component strands in alkaline CsCl equilibrium density gradients6,7. This proved that, during many-fold synthesis, the two strands in the product were not joined by an alkali stable linkage.
