Inhibition of Lysozyme by N-Acyl-D-glucosamine Derivatives

Abstract

LYSOZYME (N-acetyl muramyl hydrolase, E.C.3.2.1.17) is one of several proteins of which the three-dimensional structure is known1. The detailed mechanisms of the enzyme function and specificity are not completely established, but it is known to hydrolyse the β(1→4) glycosidic linkages, between N-acetyl-D-muramic acid and N-acetyl-D-glucosamine in the glycoaminopeptide component of the cell walls of Micrococcus lysodeikticus2 and those of chitin3. The action of lysozyme is inhibited by N-acetyl-D-glucosamine4,5 and the crystallographic structure of a lysozyme-N-acetyl-D-glucosamine complex has been described6. The activity and structure of lysozyme have been surveyed recently in a discussion organized by the Royal Society7.
