Concentration of Nicotinamide Adenine Dinucleotide in Carrageenin-induced Proliferation

Abstract

IT has been shown1 that the inhibitory effect of cortisone on the respiration of rheumatoid synovial tissue could be reversed by the addition of nicotinamide adenosine dinucleotide (NAD), and as there have been many reports2,3 of lowered co-enzyme levels in rapidly proliferating tissues, the NAD concentration of normal and proliferating synovial tissue has been determined. Proliferation was stimulated in rabbit synovia by injection into the femorotibial joint of 2 ml. of a 0.2 per cent solution of carrageenin in saline4. The synovia were removed after 1, 2, 3 or 5 days. One knee of each rabbit was injected, the other being used as control. A fairly wide range in NAD values between rabbits was found but little difference between knees of the same rabbit. NAD and NADH2 were extracted from the synovial tissue by the method of Jedeikin and Weinhouse5 and estimated by the fluorescent method using methylethylketone and alkali6.
