Evaluation of the Activity of Corticosteroid 11β-Hydroxylase

Abstract

2-METHYL-1,2-bis (3-pyridyl)-l-propanone (SU 4885) has been introduced as an agent for assessing pituitary–adrenal integrity and pituitary reserve1. The action of SU 4885 is to inhibit 11β-hydroxylation on the corticosteroid nucleus2,3. Administration of SU 4885 in the presence of a normal pituitary–adrenal relationship results in an increased excretion of 17,21-dihydroxy-20 ketosteroids as measured by the Porter–Silber1 and ketogenic steroid reactions4. This increase is a resultant of the combined effect of decreasing 11β-hydroxylation, presumably increasing adrenocorticotrophic hormone production and increasing the production of 11-deoxycorticosteroids.
